A group led by Assistant Professor Hodaka Kawai in the Department of Oral Pathology, Faculty of Medicine, Dentistry and Pharmaceutical Sciences, Okayama University, has developed a new technique to easily collect a large amount of mesenchymal stem cells that can differentiate into bone and cartilage from bone marrow tissue.

 It is known that mesenchymal cells such as bone and cartilage are not only difficult to regenerate, but also take a long time to regenerate.In addition, mesenchymal stem cells, which differentiate into mesenchymal cells, are present in only a small number in tissues and are difficult to collect, so transplantation is considered difficult.

 In this study, we developed a method that can easily isolate and extract a large amount of mesenchymal stem cells from bone marrow tissue in just 10 minutes.They discovered that if the bone marrow tissue collected from the femur and fibula of experimental mice was treated with an enzyme for 10 minutes, a significantly larger amount of mesenchymal stem cells could be collected than without the enzyme treatment.

 The collected cells can be transplanted, and as a result of observing mice transplanted with enzyme-treated bone marrow, many osteoblasts, chondrocytes, and stromal cells were observed even after 2 months after transplantation.On the other hand, these were not observed in mice transplanted with bone marrow without enzymatic treatment.In other words, not only can a large number of mesenchymal stem cells be collected by a technique that enzymatically treats bone marrow tissue for 10 minutes, but after transplantation, the bone marrow-derived mesenchymal stem cells can engraft in vivo and continue to grow for a long time.  In the future, if it becomes possible to apply this technology to collect mesenchymal stem cells from bone marrow tissue and transplant them into humans, it may be possible to promote healing of bone fractures and regeneration of bones.The results of this research are expected to lead to the establishment of next-generation regenerative medicine for patients with bone fractures that take a long time to heal, such as the elderly, and for patients with osteogenesis imperfecta who are born with bone hypoplasia.
